Immunofluorescence staining of HEp-2 cells by a representative recombinant antibody cloned from a mature naive B cell of a patient with IPEX (immune dysregulation, polyendocrinopathy, enteropathy, X-linked) syndrome. The staining shows a cytoplasmic pattern, revealing antibody autoreactivity. The frequency of autoreactive clones is increased in mature naive B cells from IPEX patients who harbor nonfunctional regulatory T cells due to mutations in the forkhead box protein 3 (FOXP3) gene.
