Overlay of a model of the type I CD20 antibody rituximab (red) and the type II CD20 antibody GA101 (blue) bound to a model of the CD20 membrane protein (white) based on the crystal structure of GA101 Fab in complex with a cyclic CD20 peptide (cyan) and on the crystal structure of rituximab-Fab in complex with the same cyclic CD20 peptide (yellow; Du J, Wang H, Zhong C, et al. Structural basis for recognition of CD20 by therapeutic antibody rituximab. J Biol Chem. 2007;282(20):15073-15080). The comparison of the two crystal structures shows that rituximab and GA101 recognize an overlapping epitope (green) on the large loop of CD20 but bind to this epitope in a completely different orientation. Note that rituximab and GA101 cannot bind to CD20 simultaneously. This composite model was prepared by Wolfgang Schaefer and Guy Georges using the Insight/Discover software package from Accelrys Inc, San Diego. See the article by Niederfellner et al on page 358.
