The cover image shows two baby hamster kidney–derived (BHK) cells expressing the hemophilia A–associated factor VIII A3 domain mutant N1922S (top) and 2 BHK cells expressing wild-type factor VIII (bottom). Factor VIII is detected with an anti–factor VIII murine monoclonal antibody, 2-54, and DyLight 488 donkey anti–mouse antibody (pseudocolored green). The Golgi apparatus is localized using wheat germ agglutinin–Alexa Fluor 350 conjugate (pseudocolored red). Nuclei are stained with DRAQ5 (pseudocolored blue). Factor VIII colocalizes with Golgi in wild-type cells (red-green overlap, yellow), but not in N1922S cells, demonstrating a block in transport of N1922S factor VIII from the endoplasmic reticulum to Golgi.
